[от имен А. Берка и П. Шарпа; греч. methodos — способ познания]метод картирования интронов и экзонов в структурном гене с использованием нуклеазы S1, которая специфически расщепляет одноцепочечную ДНК. Первоначально зрелую мРНК гибридизуют с анализируемым геном, гибрид обрабатывают нуклеазой S1 и после удаления РНК из РНК / ДНК-гибрида получают фрагменты ДНК, соответствующие экзонам этого гена.
